What Is SPRAVATO®?

SPRAVATO is a nasal spray formulation of esketamine, derived from ketamine and approved for use in patients who haven’t found relief with traditional antidepressants. Administered in-clinic only under strict observation, SPRAVATO can produce rapid improvements in mood and symptoms.

Is Spravato ketamine?

Yes. Ketamine exists as a mixture of two mirror-image molecules, known as the S- and R-enantiomers. SPRAVATO contains only the purified S-enantiomer (esketamine), which is FDA-approved for treatment-resistant depression and administered as a nasal spray.

How Does SPRAVATO Work?

SPRAVATO targets NMDA receptors in the brain, triggering increased neural connectivity and rapid antidepressant effects. Unlike SSRIs, which can take weeks, SPRAVATO may begin to work within hours or days.

How long does Spravato last?

Many patients experience improvements that last days to weeks, especially when paired with psychotherapy.

What is ketamine nasal spray used for?

SPRAVATO is used to treat treatment-resistant depression and depression with suicidal thoughts in adults.

SPRAVATO REMS: Why In-Clinic Treatment Matters

SPRAVATO is not available for home use. It must be administered in a REMS-certified clinic like Klarisana to ensure:

  • Monitoring for side effects (dissociation, blood pressure changes)

FAQ

SPRAVATO® is a prescription medicine used with or without an antidepressant taken by mouth, to treat adults with treatment-resistant depression (TRD) with an antidepressant taken by mouth, to treat depressive symptoms in adults with major depressive disorder (MDD) with suicidal thoughts or actions.

Typically, sessions occur twice a week initially, then adjust based on individual response and provider guidance.

Most patients will start with a 56mg dose of Spravato, and increase to 84mg dosing by the second session

Once in the maintenance phase, if it is determined to be appropriate, patients can move back down to the 56mg dose

Clinical trials show that SPRAVATO® can be highly effective for treatment-resistant depression. In one Phase 3 study, 69.3% of patients receiving SPRAVATO plus an oral antidepressant responded (with a 50% or greater reduction in depression scores), compared to 52% for placebo. Remission rates were 52.5% for SPRAVATO versus 31% for placebo. More recent data from the ESCAPE-TRD study also found that patients were 1.5 times more likely to achieve remission at 8 weeks when using SPRAVATO compared to quetiapine XR.
